The mechanism: hydrolyzed collagen breaks into dipeptides and tripeptides that are absorbed and seem to signal collagen synthesis in target tissues.

The question for this community: does oral collagen supplementation affect penile tissue specifically?

Honest answer: there's no research on this specifically. The general mechanism of action doesn't discriminate by tissue type in theory — collagen-producing fibroblasts exist throughout the body including penile tissue. Topical creams — what do they actually do and which ones are worth anything?

Spent about a year testing various topical products including VigRX Oil, Prosolution Gel, various alprostadil creams, and some peptide-based topicals. Here's what I found. Vasodilating topicals — particularly those with alprostadil or nitroglycerin — produce a real, fast, temporary engorgement effect. This is the same mechanism as the pills but more direct and faster acting. Erection quality and temporary fullness improve noticeably within minutes. Duration 1-2 hours. Herbal blend creams — mostly nothing. The absorption through skin is limited and the active concentrations are too low. Collagen-stimulating peptide topicals — more interesting and I'll post separately about those.

There's a lot of enthusiasm in peptide communities and a lot of motivated reasoning. Here's a framework I use for evaluating any peptide claim for enhancement:

Is there a plausible biological mechanism? (Does the peptide interact with receptors or pathways that could produce the claimed effect in the target tissue?)

What's the evidence quality? (Cell culture

Is the target tissue actually responsive? (A peptide that causes muscle growth doesn't automatically cause penile tissue growth — different receptor expression.)

What are the delivery challenges? (Topical absorption, peptide degradation in GI tract, local vs systemic distribution all affect whether the active compound reaches the target tissue.)

What's the risk profile? (GHK-Cu topical: very safe. IGF-1 systemic: significant risk. Know what you're working with.)

Apply this framework and most enhancement peptide claims survive step 1 but struggle at step 2 or 3.

Let me make the most honest biological case I can for collagen-based approaches to penile girth. This is where the science is closest to mechanistically plausible:

The penis contains significant collagen — particularly in the tunica albuginea (the fibrous sheath surrounding the corpus cavernosum) and in the connective tissue throughout the shaft. This collagen is produced by fibroblasts that express receptors for collagen-stimulating peptides including GHK-Cu.

If collagen-stimulating peptides produced meaningful additional collagen deposition in penile tissue over time, this would theoretically increase girth — because more tissue mass = more circumference. The glans specifically has a high collagen content and might respond to collagen stimulation in a way that produces visible thickening.

The problem: we don't know if the collagen response in penile tissue is meaningful, how much collagen deposition is needed to produce measurable girth change, or what the time course looks like. This is biologically plausible but unproven.

I want to give a fair overview of the actual science here because these get lumped in unfairly with snake oil pills. L-arginine is a precursor to nitric oxide. More NO means vasodilation — blood vessels relax and expand. L-citrulline is more bioavailable and converts to arginine in the kidneys. The clinical evidence for erection quality improvement is real and replicated. Studies show meaningful improvement in erectile function scores for men with mild to moderate ED. What this does NOT do: change penile anatomy, increase structural size, or produce any lasting physical change. The effect is present only while the supplement is active in your system.
